ABSTRACT

The objectives of this study were to propose a statistical model to predict the behaviour of the thickness of the uteroplacental junction as a function of the gestation period in female dogs and to determine the relationship between the thickness of the placenta and gestational age in healthy female dogs whose pregnancies had elapsed without maternal-fetal alterations. Eight Border Collie female dogs were selected, aged 3-6 (4.48 ± 0.89) and weighing 16-22 kg (19.06 ± 1.9 kg). Female dogs with gestational ages from 20 to 62 days were examined weekly using B-mode ultrasonography. Ultrasound measurements of the uteroplacental junction were organized into four distinct groups: GT1 (27-36 days of gestation), GT2 (37-46 days of gestation), GT3 (47-56 days of gestation) and GT4 (57-62 days of gestation). Based on multiple linear regression, a statistical model was proposed to predict the behaviour of the thickness of the uteroplacental junction (y) as a function of the length of gestation (x) in female dogs, where b0 is the intercept (linear coefficient) and bp is the slope of the predictors. The analysis relating GT, age and weight could predict placental thickness and resulted in a statistically significant model [F(1,28) = 153,736; p < .001; R2 = .846], but only that relating the length of pregnancy (ß = .92; t = 12.399; p < .001) predicted the thickness of the placenta according to the equation y = b0 + bp.x1 [(thickness in cm) = ß -0.3 + 0.019 × (gestation time in days)]. Only in GT4 was there no correlation between placentas within the same pregnancy (p > .05). Based on the close relationship between the development of the uteroplacental junction thickness during pregnancy and gestational age, it is possible to develop a new tool to complement gestational ultrasound evaluation in female dogs. This is important because it allows better placental evaluation in the search for significant alterations that could compromise maternal-fetal health.

ABSTRACT

A 12-year-old Saanen goat presented with a history of hyporexia and sudden recumbency. Euthanasia was indicated due to suspicion of hepatic neoplasia associated with senility. Necropsy revealed generalized oedema and increased liver size and weight (33 × 38 × 17 cm and 10.6 kg, respectively), with a firm, multilobular mass. Fusiform to polygonal neoplastic cells, with marked pleomorphism, anisocytosis and anisokaryosis, were seen on histopathological examination of the hepatic mass. The neoplastic cells were immunohistochemically positive for alpha-smooth muscle actin and vimentin but immunonegative for pancytokeratin. The Ki-67 index was 18.8%. A poorly differentiated leiomyosarcoma was diagnosed on the basis of the gross, histopathological and immunohistochemical findings and should be included in the differential diagnosis of liver disease in goats.

ABSTRACT

A 7-year-old female goat developed progressive ataxia, which progressed to sternal recumbency. Clinical examination revealed a slight elevation in the sacral spine (S1-S2), tetraparesis, perineal hypoaesthesia and absent tail and anal reflexes. Due to unresponsiveness to treatment with corticosteroids, the goat was euthanized. At necropsy, a 4.0 × 5.7 × 2.5 cm tumour mass was found in the uterine body and right uterine horn. In the cauda equina (L6‒S2), a 3 cm tumour associated with a 2 cm ventral intraosseous sacral haematoma was also found. The tumours were characterized by neoplastic proliferation of columnar epithelial cells with a predominant tubular pattern. Neoplastic cells with glandular cytoplasm stained with acid Alcian blue and periodic acid‒Schiff. Other metastases were found in the lungs, right ovary, dura mater and nerve roots of the medullary cone. Neoplastic cells were immunolabelled for cytokeratin but were negative for vimentin, and the tumour was diagnosed as metastatic endometrial tubular adenocarcinoma. To our knowledge, this is the first report of intramedullary metastasis to the spinal cord of this tumour in any species except humans.

ABSTRACT

The aim of the study was to correlate the spectral index of the right and left uterine arteries with equine placental development in mares with advanced pregnancies. We examined 32 multiparous Mangalarga Marchador mares with gestation of 150-240 days. During pregnancy, the pulsatility index (PI) and resistance index (RI) of the uterine arteries were obtained using spectral Doppler ultrasonography, and the combined uteroplacental thickness was obtained monthly using B-mode ultrasonography. The combined uteroplacental thickness correlated with gestational time of up to 13 years of age, and the significant difference was observed from the sixth month onwards. The CUPT stayed within the ideal measurements for this breed and for this gestational period. There was no correlation of CUPT with PI, however a negative and significant correlation of the RI was observed. The resistance index differed significantly among age groups, and the RI of the left uterine artery tended to decrease in all age groups. Furthermore, only RI differed significantly between the medians of gestatinal age. The left PI dropped in older mares. Thus, there an increased blood perfusion in the uterine arteries of mares with advanced pregnancies and among different age categories during placental and fetal physiological development.

ABSTRACT

Sporotrichosis is a chronic fungal infection caused by Sporothrix species. The occurrence of cases that are resistant to long-term treatment, especially in the nasal planum of cats, emphasizes the importance of studying its pathogenesis. The purpose of this study was to analyze and compare the inflammatory process of cutaneous lesions of feline refractory sporotrichosis to clinical aspects through cytopathological and histopathological examination. Moreover, the study included 13 cats with cutaneous lesions that had been resistant to itraconazole treatment for more than a year. Cutaneous lesions samples were collected for cytopathological, histopathological, and fungal culture analyses. Tissue fragments were processed and stained with hematoxylin-eosin (HE) and Grocott methenamine silver (GMS). Further, two clinical presentations had the highest occurrence: the localized cutaneous form in animals with good general condition and stable disease (n=9, 69.2%) and the disseminated cutaneous form in cats with poor general condition (n=4, 30.8%). In cats with refractory sporotrichosis, the nasal planum (84.6%) was the most common location of lesions. In the cytopathological study, cats with fewer than two lesions and in good general condition (n=9, 69.2%) showed absence or mild yeast intensity (up to 5 yeasts per field), lower intensity of macrophages and neutrophils, and higher intensity of epithelioid cells, lymphocytes, plasma cells, and eosinophils. On the other hand, (n=4, 30.8%) of the cats with disseminated sporotrichosis and a poor general condition had a marked intensity of yeasts, which were mostly phagocytosed by an increased number of macrophages and neutrophils. Of those animals with good general condition, the majority (n=6, 66.7%) had higher eosinophil intensity. In histopathology, malformed suppurative granuloma was the predominant type (n=9, 69.2%) in feline sporotrichosis lesions, followed by well-formed granulomas (n=4, 30.8%). Malformed granulomas showed mild to moderate fungal intensity (55.6%) in animals with good general condition and localized lesions while marked fungal intensity (44.4%) in cats with the disseminated form of the disease and poor general condition. Well-formed granulomas (n=4, 30.7%) had mild to moderate intensity of fungal load, and 75% of the animals with this type of granuloma had only one lesion and were in good general condition. Long-term itraconazole treatment in these cats with refractory sporotrichosis can keep the infection under control and localized lesions stable; however, fungus reactivation can occur, resulting in an exuberant and inefficient immune response.(AU)

ABSTRACT

The yellow-breasted capuchin (Sapajus xanthosternos) and robust tufted capuchin (Sapajus robustus) are endangered species due to destruction of their natural habitat and predatory chase. However, it is still necessary to elucidate some details of their reproductive physiology in order to obtain better indices in the assisted reproduction of these species. This study aimed to evaluate the ovarian cycle of 13 dominant and subordinate females of S. xanthosternos (n=8) and S. robustus (n=5) using sagittal and transversally scanned ultrasound of their uterus and ovaries. Sonograms were performed every seven days for two months. The ovarian cycle phase and anestrous condition were confirmed by colpocytology. Our results showed different uterine parameters (craniocaudal diameter, dorso-ventral diameter, and transverse diameter) (P<0.05) between anestrous subordinate females and other ovarian cycle phases and social classes. The mean of uterine volume was higher in dominant females than subordinate females in all cycle phases (P<0.05), except in follicular phase. During anestrus, endometrial width was smaller in subordinate females than in dominant females (P<0.05). Subordinate females showed differences in endometrial measures (P<0.05) between anestrous period and follicular and luteal periods. Ovarian measures in dominant females were higher than in subordinate females only during anestrus (P<0.05). In the subordinate females, ovarian parameters were different (P<0.05) between anestrus and follicular and luteal phases. Dominant females showed higher volume of right ovary compared to volume of the left ovary during anestrus and follicular phase (P<0.05). Follicles and corpus luteum were distinguished by ultrasonography in most exams (86.11%). During anestrus, measurable ovarian structures were not observed in both ovaries in dominant and subordinate females. In conclusion, the methodology used in this study allowed to evaluate the ovarian cycle in S. xanthosternos e S. robustus females and that cycle phase/anestrus and social class of the female influenced the size of the uterus and ovaries.(AU)

ABSTRACT

The aim of this study was to evaluate macro and microscopic gastrointestinal mucosa of domiciled dogs, with chronic gastrointestinal symptoms by endoscopy and histopathological evaluation, correlating clinical signs, macro and microscopy characteristics/aspects/changes of duodenal and gastric mucosa. Twenty dogs of different breeds were evaluated with with chronic characteristic signs of Inflammatory Bowel Disease (IBD), being 7 males and 13 females, with ages ranging from 1.7 to 15.8 years old. The study was prospective and cross-sectional, conducted in dogs treated at the Teaching Veterinary Medicine Hospital of the Universidade Federal Fluminense (HUVET-UFF) and private clinics in the city of Rio de Janeiro. Gastroduodenal mucosal samples were obtained by endoscopic biopsy and were processed by routine histological technique and stained with hematoxylin- eosin and Giemsa. The samples were submitted to histopathological analysis for the detection of spiral bacteria suggestive of Helicobacter spp. The rapid urease test was also performed. The clinical signs, the macro and microscopic findings were compared and correlated. In this study, we established a positive correlation between weight loss and the presence of erythema in the antrum, body and duodenum as well as correlation between change in appetite and the presence of gastric inflammatory infiltrate and positivity in the urease test. This study demonstrated the importance of the observation of some clinical signs such as weight loss and appetite changes in the course of IBD. Thus, it is emphasized that dogs showing these clinical signs should have the IBD included in the differential diagnoses group avoiding thereby neglecting a disease that increasingly affects small animals.(AU)

ABSTRACT

This study aimed to measure the physiological changes in the combined utero-placental thickness (CUPT) during the course of normal pregnancy in Mangalarga Marchador mares. Transrectal B-mode ultrasonography was used to measure CUPT in 42 pregnant mares with a mean age, 8.7 (±3.5 years). CUPT was measured every month from 150 days of gestation until delivery. CUPT was expressed as an average of three measurements of the distance (mm) between the middle part of the uterine artery and the allantoic fluid. A General Linear Model was used to evaluate the major effects of the chronological age and stage of gestation on CUPT in mares. The coefficient of determination (R2) was 0.571 (P < 0.001). There was no correlation between CUPT and the chronological age of the mares, however, there was a positive correlation between CUPT and the stage of gestation, which an accounted for 29.6% of the observed variation. CUPT significantly increased during the gestational period from 210 to 240 days and 270-300 days. These results showed CUPT increased as pregnancy advanced in the Mangalarga Marchador mares. In conclusion these effects of the stage of pregnancy on the increase in CUPT showed a physiological parameters of ultrasonography evaluation of the placenta in Mangalarga Marchador mares.

ABSTRACT

BACKGROUND: Squirrel monkeys (Saimiri spp.) have been widely used as animal models; however, the occurrence of Staphylococcus sp in their vaginal microbiota remains to be described. METHODS: Samples were collected from 175 adult squirrel monkeys to isolate Staphylococcus sp and to test for susceptibility to a panel of nine antimicrobial agents. RESULTS: Isolates with characteristics of the genus Staphylococcus were detected in 95 of 175 samples. Coagulase-negative staphylococci (CoNS) were the most common (95.8%, 91/95) isolates. Resistance to antibiotics was observed in 47.3% (45/95) of isolates. Resistance to tetracycline was observed in 28.5% (26/91), chloramphenicol in 15.4% (14/91), and methicillin in 13.2% (12/91) of CoNS. Coagulase-positive staphylococci were resistant to tetracycline, erythromycin, and methicillin. CONCLUSIONS: The presence of Staphylococcus sp in vaginal samples obtained from squirrel monkeys suggests that these animals were in a carrier state. Furthermore, isolating strains resistant to methicillin reinforces the biosafety care of a colony.

ABSTRACT

Toxoplasma gondii is a parasite considered one of the major causes of reproductive problems in sheep. Furthermore, the presence of the agent in ram semen urges the possibility of sexual transmission in this species. The aim of this study was to evaluate if ram's frozen semen spiked with T. gondii tachyzoites would be able to cause infection in sheep by laparoscopic artificial insemination (AI). Nine ewes tested seronegative to anti-T. gondii antibodies by the modified agglutination test (MAT) were superovulated and inseminated to collect embryos. Animals were divided into two groups: G1 (n = 5), ewes inseminated with semen containing 4 × 107 tachyzoites; and G2 (n = 4), ewes inseminated with tachyzoite-free semen (control group). To confirm infection, ewe's blood samples were collected on days -14, -7, 0, 7, 14, 21, 28, 35, 49 and 57 after AI for analysis by MAT and PCR. Tissue samples of these ewes were also collected for histopathology and immunohistochemistry (IHC). Seven days after AI, all ewes of group G1 had specific antibodies to T. gondii, while those of G2 were negative. Toxoplasma gondii DNA was detected in the blood of one ewe and parasites were observed in tissues of all five animals inseminated with contaminated semen, indicating that semen freezing protocol does not affect T. gondii transmission by artificial insemination in sheep.

ABSTRACT

Vinte e seis animais foram inclusos no Grupo I e vinte e cinco no Grupo II.

Hyperthyroidism is the most common endocrine disorder of older cats. Due to the progressive nature of the disease, the identification of a subclinical stage is essential in order to have a better control of the disease. The present study was a survey of the frequency of feline hyperthyroidism in Rio de Janeiro, from March, 2007 to April, 2008. The purpose of this study was to perform an evaluation of the clinical, laboratorial and echocardiographic aspects in a feline population of naturally acquired hyperthyroidism, with or without palpable thyroid gland. The selection of the cats was done by the documentation of an elevation of total thyroxine (TT4) levels by radioimmunoassay. The animals were divided in two groups according to the thyroid palpation: group I (non-palpable thyroid lobe) and group II (palpable thyroid lobe). Laboratorial and complete clinical evaluations were performed in all cats. The clinical evaluation included behavioral evaluation, body weight, body condition, dermatologic, cardiopulmonary auscultation, thyroid palpation and systolic blood pressure reading. Besides the TT4, the following exams were performed: urea, creatinine, alanine aminotransferase (ALT), alkaline phosphatase (ALP) and glucose. Thirty one cats had an echocardiogram performed. On the basis of TT4 concentrations, 51 of the 178 cats were classified as hyperthyroid. One or more enlarged thyroid glands were palpated in 25 of the 51 hyperthyroid cats and were included at Group II. Twenty six cats didn't palpable thyroid gland and were included at Group I. Clinical, laboratorial and echocardiographic parameters were similar between the two groups. However, the TT4 levels were significantly lower in the non-palpable thyroid group. All geriatric cats must be thyroid gland palpable on clinic examination and have TT4 measured. Many hyperthyroid cats may be asymptomatic and early diagnosis prevents organ damage such as liver, kidneys and heart.

ABSTRACT

Extracellular matrix (ECM) components such as fibrillar collagens play a fundamental role in wound repair and have also been studied in association with the gastric ulcer healing process in gastroenterology. Nevertheless, there have been no studies in the literature to date regarding the description and characterization of ECM components, neither in normal nor in injured gastric tissue of primate species. The objective of this study was to investigate the expression of gastric collagen types I, III, and IV in marmosets (Callithrix sp.). Histological specimens from the stomach of 6 Callithrix jacchus, 12 C. kuhli, and 12 C. geoffroyi were evaluated. The specimens were immunostained with anti-types I and III collagen polyclonal antibodies and anti-type IV collagen monoclonal antibody. Collagen types I and III were detected in the submucosa and lamina propria between the mucosal glands while collagen type IV was detected in the muscularis mucosae, muscular layers, blood vessels, and gastric mucosa between the mucosal glands. It is hoped that these findings can contribute to future studies on the gastric extracellular matrix components in primates and to comparative studies in the area of gastroenterology.

ABSTRACT

The vast majority of mammary tumors in cats are malignant with high metastatic and mortality rates. This study evaluated clinical, cytological and histopathological aspects of 20 cases of cat mammary masses deriving from the city of Rio de Janeiro. Data on history, macroscopic description, evidence of regional lymph node and distant metastases, clinical examination and treatment were collected. The cats were treated with unilateral mastectomy, unilateral mastectomy plus chemotherapy or chemotherapy alone. Tissues were submitted for cytological and histological evaluation. Histopathological analysis described one case of mammary hyperplasia and 19 cases of adenocarcinoma. Inadequate surgical margins were obtained in two cats while six showed evidence of microscopic invasion in other mammary glands. Multiple mammary involvement and greater tumor size (diameter over 7.05 cm) were correlated with increased chance of local metastases. Ulceration of tumors and metastases were common. These facts support the highly malignant nature of this neoplasia and emphasize the risks associated with delay in diagnosis and treatment of feline patients. In conclusion, mammary palpation should be performed routinely in the clinical practice.
